SEQUENCING WORKORDER

 

PLEASE READ CAREFULLY:
Fill this form out completely each time you submit a sample. Multiple samples with the same primer(s) maybe be listed in chart format on the lower right or on the back of the sheet (don’t forget to include a concentration for each sample). All sample tubes must be labeled clearly with the template name on the top. On the side of the tube include the name of the advisor (P.I.) and the concentration. Sample names MUST be eight characters or less, and primer names MUST be six characters or less. All samples and primers need to be resuspended in water. Plasmid concentrations should be 20ng/ul to 200 ng/ul, please provide a minimum of 200ng of plasmid template for each requested primer. PCR product concentrations should be 10 ng/ul to 30 ng/ul, please provide a minimum of 30ng of template for each requested primer. Primers must be supplied at 3.2 pmol/ul (see bottom of page).

DATA OUTPUT AND STORAGE: Paper copies of data are no longer provided, you will receive electronic copies via email or PSI server (discuss the set up process with us). Keep in mind that the data files are large so we recommend using storage media such as zip disks, cds, or dvds. In addition, the sequencing facility will continue to archive your data.

PRIMER(S) TO USE FOR SEQUENCING:

 

To make a 3.2 pmol/ul working solution of your primer use the rule "n+2"ug/ml, where n= the length of the primer in nucleotides. e.g. If your primer is 17 nucleotides long add 2 to give 19, then dilute an aliquot of your primer stock to 19 ug/ml in water. This will give a solution that is approximately a 3.2 pmol/ul.
